Calcium signal is regulated by the stiffness of the substrate presenting antigen
As the "second messenger", calcium plays an important role in signal transduction pathway in a diversity of cells, including B lymphocytes. B cells get activated after B cell receptor`s encountering antigen, followed by calcium signaling initiation, which represents the level of activation of B cells. Antigen is always presented on substrates of different stiffness in vivo, and B cells response in different ways depending to the stiffness. However, how antigen stiffness affect calcium signal in B cell is still unknown. To answer this question, confocal microscopy is applied to collect calcium signal image data from B cells which have recognized antigen coated on substrate with different stiffness. The results show that, antigens on stiffer surface induce higher frequency of occurrence of calcium oscillation, more swift amplitude attenuation, but shorter duration of single calcium oscillation.
