Study on Pluripotency Variation During Early Culture of Zebrafish Embryo Cells
In this study, the expressions of pluripotency genes were examined at the initial stage of the cultured zebrafish middle blastula/gastrula cells. In the cultured blastula cells, the expression of pluripotency genes can be classified into 4 groups, and the pluripotency mechanisms of zebrafish may be different from mammals like mouse and human, since the core pluripotency genes like Pou5f3/Oct4 and Nanog are declined 1000 fold with the blastula cells cultured in vitro. In the cultured gastrula cells, most of the pluripotency genes are declined for at least 50℅ after 3 days culture but for Pou5f3/Oct4, Nanog and Sox2 with a short up-regulate and then declines. The expression of Sall4 and Klf4 are declined both in blastula/gastrula cultured cells, adult somatic cells and long term cultured blastula cells as compared to uncultured blastula cells, so Sall4 and Klf4 may be a suitable stemness marker in zebrafish ES cells culture. This work laid a foundation for clarifing pluripotency mechanisms of zebrafish and the establishment of diploid/haploid zebrafish ES cells which can stably self-renewal and maintain high pluripotency for long term cultured in vitro.
